摘要

Abstract

This study was conducted to develop Campylobacter coli standards using droplet digital PCR(ddPCR)detection method.The glyA gene was used as the target gene to establish the ddPCR method,and the specificity,reproducibility and sensitivity of the method were tested and clinically applied.Candidate standards containing the Campylobacter glyA gene were prepared,and the established ddPCR method was applied to evaluate the homogeneity and stability of the standards,and the values were jointly determined by nine laboratories.The established dd PCR method has high specificity,good reproducibility and high sen-sitivity;the prepared standard substance has good homogeneity and can be stably stored for 12 months at-20℃,and the result of the calibration was 5202.60 copies/μL with an uncertainty of 0.43 × 103 copies/μL.The development of this standard substance provides the molecular biology detection method of Campy-lobacter colonis,and provides the support of metrological traceability and quality control for the molecular biology detection method of Campylobacter coli,and helps to promote the standardisation of related detec-tion reagents.
