Study on the Synergistic Antibacterial Effect of Paeoniflorin Combined with Zinc Hyaluronate against Staphylococcus pseudointermedius in Vitro

Abstract

This study aimed to investigate the in vitro antibacterial effects of the combined use of paeoni-florin and zinc hyaluronate against Staphylococcus pseudointermedius,a major pathogenic bacterium re-sponsible for bacterial skin infections in pets,and to explore the underlying antibacterial mechanisms.The in vitro antibacterial efficacy was determined by measuring the diameter of inhibition zones,growth inhibi-tion curves,viable bacterial counts,and bacterial inhibition rates following treatment with paeoniflorin,zinc hyaluronate,or their combination.Additionally,the antibacterial mechanisms were explored based on the effects of these treatments on biofilm formation,leakage of intracellular macromolecules such as nucleic acids and proteins,and morphological changes in bacterial cells.The results showed that,compared to the use of paeoniflorin and zinc hyaluronate alone at 2.5 mg/mL,their combined use significantly increased the inhibition rate against Staphylococcus pseudointermedius and the leakage of nucleic acids and proteins,while significantly reducing the number of viable bacteria and the biofilm formation rate(P<0.05).Moreo-ver,the combination treatment induced severe morphological alterations in bacterial cells,including pro-nounced shrinkage,collapse,and damage.Therefore,under the conditions of this experiment,the combined use of paeoniflorin and zinc hyaluronate exhibited significantly stronger antibacterial effects against Staph-ylococcus pseudointermedius than that when used individually.The mechanism of this synergistic antibac-terial activity is primarily achieved through the disruption of biofilm formation,induction of intracellular nucleic acid and protein leakage,and inhibition of normal bacterial growth.
