波罗蜜花粉生活力测定与花粉萌发培养基的优化
Determination of pollen viability and optimization of pollen germination medium in Artocarpus heterophyllus Lam

Abstract
[Objective]This study aimed to screen the optimal medium formulation for in vitro germination of Artrocarpus heterophyllus Lam.pollen and to evaluate the applicability of rapid staining method for detecting pollen viability.[Method]Pollen of'Siji'A.heterophyllus was used as experimental material to measure pollen germination rate and optimize pollen culture medium composition by single factor test and orthogonal test.[Result]The effects of sucrose,H3BO3,CaCl2 and MgSO4 on pollen germination rate were significant,with CaCl2 ranking first,followed by sucrose,H3BO3,and MgSO4.The optimum medium formula was 5%sucrose+150 mg/L H3BO3+100 mg/L CaCl2+150 mg/L MgSO4,and the germination rate was 72.39%,which was significantly higher than other treatments.The optimum germination temperature was 27℃.Both the MTT staining method(10 min staining,62.99%viability)and the TTC staining method(45 min staining,78.58%viability)could be used for rapid detection of pollen viability.The MTT staining method was more efficient and had better contrast ratio while the magenta acetate and I2-KI methods were not applicable.[Conclusion]The results provide a theoretical basis and practical support for pollen viability determination and artificial pollination technology in A.heterophyllus.cross breeding.关键词
