基于SRAP分子标记的石斛兰杂交后代鉴定及其遗传多样性分析
Identification and genetic diversity analysis of Dendrobium hybrid progeny based on SRAP markers

Abstract
[Objective]The authenticity identification method of Dendrobium hybrid progeny based on SRAP marker technology was constructed to provide reliable technical support for the identification of new varieties(lines)of Dendrobium.[Method]Using SRAP marker technology,the authenticity identification and genetic diversity analysis of D.primulinum,D.anosmum and 151 hybrid progeny which were from D.primulinum as female parent and D.anosmum as male parent were carried out.[Result]7 pairs of specific primers were screened from 90 pairs of SRAP primers to achieve efficient identification for hybrid progeny,with an efficiency of 100%;Polymorphism analysis showed that the polymorphism ratio of 7 pairs of primers was 81.03%,revealing rich genetic variation;Cluster analysis showed that 153 sample materials could be divided into five groups when the genetic distance was 0.24.Among them,82.78%of the hybrid progeny were clustered with male parent,and the genetic relationship was closer.The genetic similarity coefficient among the hybrid progeny lines was generally higher than that between them and the two parents,and the genetic similarity coefficient between the male parent and the hybrid progeny lines was close to that between the female parent and the hybrid progeny lines.[Conclusion]SRAP marker can be used for the early identification of the hybrid progeny of Dendrobium,which can effectively assist the identification and breeding of new varieties.关键词
