Abstract

In order to screen the optimal medium formulation for callus induction from leaves of Camellia fascicularis,the leaves of tissue-cultured seedlings of Camellia fascicularis were used as the materials,the effects of hormones,light conditions and placement methods on the callus induction of leaves were discussed.The formula of callus induction medium was optimized through the orthogonal experiment.The results showed that the methods of contacting the blade back of the leaf with the medium and maintaining the complete darkness during the whole process of cultivation were most conducive to the callus induction.The optimal medium formulation for leaf callus induction was 1/2MS+45 g·L-1 sucrose+3.0 mg·L-1 NAA.This study established the technical system for callus induction from the leaves of Camellia fascicularis,which laid a preliminary foundation for the subsequent development of genetic transformation system by using callus.
