太子参灰霉病拮抗放线菌的分离与鉴定
Isolation and identification of antagonistic actinomycetes against gray mold in Pseudostellaria heterophylla

Abstract
[Purpose]The study isolated and screened actinomycetes strains with antagonistic activity against Botrytis cinerea from the rhizosphere soil of Pseudostellaria heterophylla,aiming to provide a reference for effectively preventing gray mold caused by B.ci-nerea in P.heterophylla.[Method]With B.cinerea as the indicator fungus,actinomycetes were isolated from the rhizosphere soil of P.heterophylla via the plate dilution method.The antagonistic activity of each isolate against B.cinerea was determined by the stand-ard plate confrontation assay,and the strains exhibiting significant antagonistic effects were selected.Their taxonomic status was i-dentified based on morphological characteristics,physiological and biochemical properties,and molecular biology(16S rDNA se-quencing).[Result]A total of 48 actinomycetes strains were isolated from the rhizosphere soil of P.heterophylla.Among them,strains E-6,E-3,A-2,A-5,B-10,and C-2 exhibited inhibitory effects against B.cinerea.Primary screening results indicated that strain E-6 demonstrated the best plate antagonism activity,forming an inhibition zone with a diameter of 14.25 mm.Secondary screening results showed that the fermentation broth of strain B-10 had the strongest antagonistic effect,achieving an inhibition rate of 47.4%on day 4 of culture.Strains B-10 and E-6 were identified as members of Streptomycetaceae.Specifically,strain B-10 was identified as a member of Kitasatospora,showing the 16S rDNA sequence similarity of 98.92%compared with K.xanthocidica.Strain E-6 was identified as a member of Streptomyces,presenting the 16S rDNA sequence similarity of 99.59%compared with S.atratus.[Conclusion]Strain B-10,identified as K.xanthocidica,can serve as a candidate biocontrol strain for the research on antibiotics.Strain E-6,identified as S.atratus,can be considered a candidate strain for research into biocontrol microbial fertilizers.Both strains exhibit potential for development as biocontrol agents against gray mold in P.heterophylla.关键词
