脑胶质瘤中KLRB1和PRDX1的表达及其对预后的预测价值
Expression of KLRB1 and PRDX1 in glioma and their prognostic predictive value

Abstract
Objective:Glioma is a common malignant tumor of the central nervous system.Identifying biomarkers that can effectively assess glioma prognosis is essential for patient stratification.Killer cell lectin-like receptor B1(KLRB1)and peroxiredoxin 1(PRDX1)have been found to be upregulated in various malignancies.This study aims to investigate the expression of KLRB1 and PRDX1 in glioma tissues and analyze their prognostic value. Methods:A retrospective analysis was conducted on glioma patients diagnosed by pathology at Chengdu Shangjin Nanfu Hospital/West China Hospital,Sichuan University Shangjin Hospital from February 2019 to February 2021.The mRNA and protein expression of KLRB1 and PRDX1 in glioma and adjacent tissues were detected using real-time fluorescence quantitative PCR and immunohistochemistry.Pearson correlation analysis was used to assess the correlation between KLRB1 and PRDX1 mRNA expression.The association of KLRB1 and PRDX1 protein positivity with clinicopathological parameters was analyzed.Kaplan-Meier survival analysis and the Log-rank test were used to compare survival curves,and a Cox regression model was used to identify prognostic factors. Results:The mRNA expression levels of KLRB1 and PRDX1 in glioma tissues were 3.12±0.45 and 2.67±0.42,significantly higher than those in adjacent tissues(0.42±0.09 and 0.68±0.12)(KLRB1 t=58.244,PRDX1 t=45.100;both P<0.001).KLRB1 and PRDX1 mRNA levels were positively correlated(r=0.712,P<0.001).The positive expression rates of KLRB1 and PRDX1 proteins in glioma tissues were 63.27%(62/98)and 65.31%(64/98),significantly higher than in adjacent tissues(6.12%and 8.16%)(KLRB1 χ2=70.618,PRDX1 χ2=68.846;both P<0.001).Gliomas≥3 cm in diameter and WHO grade Ⅲ-Ⅳ showed significantly higher KLRB1 and PRDX1 positivity compared to tumors<3 cm and World Health Organization(WHO)grade Ⅰ and Ⅱ(P<0.05).The 3-year overall survival rate was significantly lower in the KLRB1-positive group(27.42%)and PRDX1-positive group(25.00%)than in the KLRB1-negative(55.56%)and PRDX1-negative(61.76%)groups(KLRB1:Log-rank χ2=6.439,P=0.011;PRDX1:Log-rank χ2=11.010,P=0.001).WHO grade Ⅲ-Ⅳ,tumor diameter≥3 cm,and positive expression of KLRB1 and PRDX1 were identified as independent prognostic factors(all P<0.001). Conclusion:KLRB1 and PRDX1 are upregulated in glioma tissues,and their positive expression is independently associated with poor prognosis in glioma patients.These markers may serve as useful indicators for prognostic evaluation.关键词
