基于Indel分子标记的分蘖芥菜种质资源遗传多样性及指纹图谱构建
Genetic Diversity and Fingerprinting Construction of Tillering Mustard Germplasm Resources Based on Indel Molecular Markers

Abstract
[Objective]The genetic diversity and genetic relationship of different tillering mustard germplasms resources were explored to establish a technical system for identification of different tillering mustard inbred lines and provide the theoretical basis for improving mustard varieties and speeding up the breeding process.[Method]The phenotypic characters of 22 tillering mustard germplasm resources(B1-B23)with abundant genetic diversity were statistically calculated.The fingerprint chromatograms of 22 tillering mustard inbred lines were drawn by 72 pairs of Indel markers developed from the sequencing results to analyze their genetic relationships and genetic distance.[Result]22 tillering mustard germplasm resources were clustered into two groups morphologically when the Euclidean distance was 20.When the Euclidean distance was 15,22 tillering mustard germplasm resources were clustered into Group A(B2,B18,B10,B15 and B22),Group B(B1,B19,B5,B9,B23,B6,B20,B17 and B8)and Group C(B14,B11,B16,B4,B7,B3,B21 and B12)morphologically and there were great differences in leaf type and leaf margin crack among three groups.The genetic similarity coefficients of 22 tillering mustard inbred lines were 0.58-0.99.When the genetic similarity coefficient was 0.64,22 tillering mustard inbred lines could be clustered into Subgroup A(B22),Subgroup B(B4,B6,B8,B9,B14 and B20),Subgroup C(B3,B12,B16,B17 and B21)and Subgroup D(B1,B2,B5,B7,B10,B11,B15,B18,B19 and B23).6 pairs of core primers were screened by Indel markers and the fingerprint chromatograms of 22 tillering mustard inbred lines were drawn.[Conclusion]The fingerprint chromatograms of 22 tillering mustard germplasm resources are preliminarily established by investigating their botanical phenotypic traits and marker screening,which can help to identify tillering mustard varieties.关键词
