基于线粒体D-loop区和COI基因序列的毕节地区2种裂腹鱼遗传多样性分析
Genetic Diversity Analysis of Two Schizothorax Species in Bijie Region Based on Mitochondrial D-loop and COI Gene Sequences

Abstract
[Objective]The genetic diversity of Schizothorax grahami and Schizothorax kozlovi in Bijie of Guizhou was investigated to provide a reference for germplasm conservation and stock enhancement.[Method]Mitochondrial D-loop region and COI gene sequences were used as molecular markers to analyze genetic diversity of two Schizothorax species.[Result]10 and 9 haplotypes were defined based on mitochondrial D-loop and COI gene sequence respectively in the 38 individuals of S.grahami,meanwhile the haplotype diversity(Hd)and nucleotide diversity(π)were 0.781,0.630 and 0.006 44,0.003 73 respectively.5 and 4 haplotypes were defined based on mitochondrial D-loop and COI gene sequence respectively in the 34 individuals of S.kozlovi,meanwhile the haplotype diversity(Hd)and nucleotide diversity(π)were 0.524,0.485 and 0.002 62,0.001 21 respectively.The neutral detection and nucleotide mismatch distribution indicated that the two Schizothorax species may not have experienced large-scale population expansion events recently.[Conclusion]The genetic diversity of S.grahami is at a medium level in Bijie of Guizhou,whereas the genetic diversity of S.kozlovi is low.关键词
