Screening and Validation of Reference Genes in Cyclocodon lancifolius

Abstract
To investigate the optimum reference gene for gene expression analysis in different tissues and under various abi-otic stress for edible and medicinal new resource plant of Cyclocodon lancifolius(Roxb.)Kurz.In this study,five candidate internal reference genes(ACT,TUA,TUB,GAPDH,UBC)were selected based on the transcriptome data of Cyclocodon lan-cifolius.The expression levels of candidate reference genes were assessed by quantitative real-time PCR.The gene expres-sion stability analysis software was utilized to evalute the stability expression of five candidate internal reference genes of Cy-clocodon lancifolius.Subsequently,the functional genes of Cyclocodon lancifolius were used to identified and validated the optimal internal reference genes.Our findings indicated that ACT had the best expression stability in different tissues and high temperature stress,the expression stability of TUA was the best under NaCl stress,the expression stability of UBC was the best in ABA treatment and low temperature stress,the expression stability of GAPDH was the best under PEG stress.Comprehensive analysis and using SAPK1 and PP2CA as target genes to verified the screened reference genes indicated that ACT was the most suitable reference gene of Cyclocodon lancifolius in different tissues and under abiotic stresses gene ex-pression study.关键词
