Abstract

Objective:To explore the effects of Rizhong dietotherapy formula on ocular development,retinoic acid(RA)and dopamine(DA)levels and expression of related receptors in guinea pigs under different light sources.Methods:A total of 64 2-week-old guinea pigs were randomly divided into 8 groups:the light groups(natural light group,LED light group,fluorescent light group,simulated natural light group)and light diet groups(natural light diet group,LED light diet group,fluorescent light diet group,simulated natural light diet group)with 8 guinea pigs in each group.Among them,the LED group,the fluorescent group and the simulated natural light group were called the artificial light source groups.The light groups were fed with convention-al feed,and the light diet groups were fed with special feed from the Rizhong dietotherapy formula.The eye axis of each group of guinea pigs was measured every 2 weeks,the diopter was measured every 4 weeks.After 12 weeks,the morphological changes of retinal tissues were observed after H&E staining and the content of RA and DA in the retinal tissues of guinea pigs in each group was determined by high-performance liquid chromatography(HPLC).The protein and mRNA expressions of retinoic acid receptor β(RAR-β),cellular retinoic acid binding protein I(CRABP-Ⅰ)and dopamine D2 receptor(DRD2)in retinal tissues were detect-ed by Western blot and RT-PCR.Results:Compared with the natural light group,the eye axis of guinea pigs in LED and fluores-cent light groups was significantly extended,and there were statistical differences at the 4th,6th,8th,10th and 12th weeks(P<0.05).The refractive state shifted towards myopia,with differences appearing at the 4th and 8th weeks,and reaching a myopic state by the 12th week(P<0.05).The content of RA in retinal tissue of guinea pigs increased,the content of DA decreased,the mRNA and protein expressions of RAR-β and CRABP-I increased(P<0.01),and the mRNA and protein expressions of DRD2 decreased in the artificial light source groups(P<0.01).After the intervention of the Rizhong dietotherapy formula,the axial length of guinea pigs in the LED light diet group and the fluorescent light diet group was smaller than that of the LED light group and the fluorescent light group,and there were statistical differences in the 4th,6th,8th,10th,and 12th weeks(P<0.05).The diopter of hyperopia decreased slowly,which was different at the 4th and 8th weeks respectively,and was still in a hyperopic state at 12 weeks(P<0.01).Compared with the same light source illumination group,the content of RA in retinal tissue decreased,the con-tent of DA increased,the mRNA and protein expressions of RAR-β and CRABP-I decreased significantly(P<0.01),the mRNA and protein expressions of DRD2 increased significantly in light diet groups(P<0.01).The results of retinal H&E staining showed that compared with natural light group,the retinal tissue of guinea pig in the artificial light source groups was thinner,the ganglion cells were sparse and vacuolated,and the distribution gap between the inner and outer nuclear cells was large,the num-bers were reduced and the arrangement was disordered.After the intervention of Rizhong dietotherapy formula,the retinal tissue in the artificial light diet groups was thickened compared with the same light source light groups,the numbers of ganglion cells and in-ner and outer nuclear cells increased and arranged relatively neatly.Conclusion:LED and fluorescent light can cause the develop-ment of myopia in guinea pigs,and the Rizhong dietotherapy formula can reduce the level of RA,increase the content of DA in the retina of guinea pigs,regulate the expression of RAR-β,CRABP-I and DRD2,improve the tissue structure of the retina,and in-hibit the growth of the eye axis,thereby delaying the progression of myopia.
