鸡血藤提取物通过PI3K/AKT通路对缺铁性贫血大鼠的药效和作用机制研究
Efficacy and mechanism of Spatholobus suberectus extract on iron deficiency anemia rats via PI3K/AKT pathway

Abstract
Objective:To investigate the efficacy and mechanism of Spatholobus suberectus(S.suberectus)extract on iron defi-ciency anemia(IDA)rats.Methods:A low iron diet combined with cutting the tail and bleeding was used to construct an IDA rat model.The efficacy of S.suberectus extract was evaluated by measuring peripheral blood count,serum iron,total iron binding ca-pacity,transferrin saturation,and serum ferritin levels,observing morphological changes in liver and kidney tissues,and assessing the levels of oxidative stress markers and hepcidin in the liver.Furthermore,network pharmacology methods were used to screen potential pharmacodynamic targets and key signaling pathways,and the important pharmacodynamic targets in key signaling path-ways in the liver were verified by reverse transcription-real-time quantitative polymerase chain reaction(RT-qPCR)and Western blot to reveal their mechanism of action.Results:S.suberectus extract significantly improved peripheral blood count indices such as red blood cell count,hemoglobin concentration,hematocrit,and mean corpuscular volume,mean corpuscular hemoglobin,and mean corpuscular hemoglobin concentration,and slowed down the decreasing rate of serum iron and transferrin saturation in IDA rats(P<0.05),repaired the damage of kidney and liver tissues,and increasing the content of hepcidin(P<0.01)and antioxidant capacity in liver tissues.At the molecular level,S.suberectus extract upregulated the expression of PI3K and AKT mRNA,as well as the levels of PI3K,p-PI3K,AKT and p-AKT proteins.Conclusion:S.suberectus extract can promote the recovery of pe-ripheral blood count,regulate serum iron metabolism level,repair liver and kidney tissue damage,and increase hepcidin content and antioxidant capacity in IDA rats.These improvements may be achieved by activating the PI3K/AKT signaling pathway in liv-er tissues.关键词
