摘要

Abstract

Objective:To investigate the effect of Jiaji electroacupuncture at the spinal cord on the activation of microglia in rats with spinal cord injury by regulating the HMGB1/RAGE/NF-κB pathway.Methods:The SCI rat model was established us-ing Allen's method,and the rats were randomly divided into a sham surgery group,a model group,and an electroacupuncture group.BBB score was used to evaluate hind limb motor function in rats after SCI;ELISA method was used to detect the expres-sion of inflammatory factors;Western blot was used to detect the expression levels of HMGB1,RAGE,and NF-κB proteins;Im-munofluorescence was used to observe the expression of small glial cell marker Iba-1.Results:Compared to the sham surgery group,the BBB scores of the model group were significantly reduced at all time points,indicating successful modeling.The scores of the electroacupuncture group were significantly higher than those of the model group at 3,7,and 14 d.The expression of pro-in-flammatory factors in the electroacupuncture group was significantly reduced compared to the model group.The protein expression of HMGB1,RAGE,and p-NF-κB p65/NF-κB p65 in the electroacupuncture group was lower than that in the model group.The expression of Iba-1 in the electroacupuncture group was significantly reduced compared to the model group.Conclusion:Jiaji elec-troacupuncture can inhibit the activation of HMGB1/RAGE/NF-κB pathway in rats with spinal cord injury,thereby suppressing the activation of microglia,reducing neuroinflammation,and promoting the recovery of neurological function in rats with spinal cord injury.
