摘要

Abstract

Objective:A periosteum extracellular matrix(PECM)hydrogel loaded with exosomes from adipose mesenchymal stem cells(AdMSC-Exos)was prepared.And its therapeutic effects on osteoarthritis rats induced by destabilization of medial me-niscus(DMM)were evaluated.Methods:AdMSC-Exos were extracted using ultrafiltration method and then characterized.PECM were preparated using decellularization and enzymatic degradation processes.The hydrogels were loaded with exosomes to obtain PECM@AdMSC-Exo.Male SD rats were randomly divided into sham group,DMM+Saline group,DMM+PECM group,DMM+AdMSC-Exo group and DMM+PECM@AdMSC-Exo group.The therapeutic effect of the drug on osteoarthritis was observed in vivo by intra-articular injection.Results:The results of characterization experiments showed that AdMSC-Exo,PECM and PECM@AdMSC-Exo were successfully preparated.In vitro experiments showed that AdMSC-Exo had a reparative effect on injured chondrocytes.Compared with the control group,PECM@AdMSC-Exo can effectively repair cartilage and had a better therapeutic effect on osteoarthritis in rats,and had no significant effect on the body weight of rats.Conclusions:PECM@AdMSC-Exo were successfully preparated,which showed better repair effects on cartilage injury.This study provides a new strategy for the treatment of osteoarthritis.
