In vitro and in vivo evaluation of dual-loaded oxaliplatin/indocyanine green nanoparticles for the treatment of gastric cancer
摘要
Abstract
Objective:To investigate the effect of composite nanoparticles in anti-tumor therapy,and systematically evaluate the anti-tumor activity and the synergistic effect of photothermal chemotherapy of composite nanoparticles through in vitro cell and in vivo metastatic tumor model experiments.Methods:Composite nanoparticles were prepared by hot melt extrusion and freeze grinding process.Fluorescence microscopy was used to observe the color development of human epithelioid gastric cancer cells(HGC-27),and study the anti-tumor effect of nanoparticles on HGC-27 cells in vitro.The tissue sections of MKN-45 tumor-bearing nude mice were examined by fluorescence microscope under each channel to study the anti-tumor effect of nanoparti-cles on metastatic tumors in vivo.Results:The results of anti-tumor activity showed that dual-loaded oxaliplatin/indocyanine green nanoparticles(PLGA/ICG/Oxa)had the strongest inhibitory effect on tumor growth,and photothermochemotherapy com-bined with photothermotherapy and sustained release chemotherapy had a synergistic effect.Conclusion:In this study,a new PLGA/ICG/Oxa nanoparticle has been successfully prepared,which has the effects of photothermotherapy and sustained release chemotherapy,and can effectively inhibit tumor growth,providing a promising method for clinical treatment of gastric cancer.关键词
