黄淮肉羊FABP3基因组织表达及与肉质性状关系
Tissue expression and relationship with meat quality traits of FABP3 gene in Huanghuai sheep

Abstract
[Objective]The aim of this study was to investigate the relationship between FABP3 gene and intramuscular fat deposition and meat quality in Huanghuai sheep through cloning,sequence analysis,tissue expression,and correlation analysis with meat quality.[Method]3 male Huanghuai sheep at the ages of 3,9 and 18 months were selected,and samples of tissues and organs,such as semitendinosus,triceps brachii,quadriceps femoris,longissimus dorsi,rumen,subcutaneous fat,kidney,spleen,liver,lung,testis,and small intestine were collected.And the meat quality indica-tors,such as IMF content,muscle fatty acids,and quality traits after slaughter,were measured.Sub-sequently,the longissimus dorsi RNA was extracted,and the coding region sequence of the FABP3 gene was amplified and its sequence characteristics were predicted using bioinformatics analysis.The mRNA level of FABP3 in various tissues at 9 months and the longissimus dorsi at different age stages was detected by real-time quantitative PCR,and the correlation between FABP3 expression and meat quality indicators,was analyzed through Pearson correlation coefficient.[Result]The coding region size of FABP3 gene of Huanghuai sheep was 402 bp,encoding 133 amino acids,indicating its closest genetic relationship with Capra hircus.FABP3 protein was a stable hydrophilic protein with no signal peptide and transmembrane structure presence.There were 26 phosphorylation sites and the protein structure mainly contained extended chain(36.48%)and random coil(27.07%).There were interac-tions between FABP3 and ACSL1,PPARG,PLIN2,GPAM and DGAT1.The results of Real-time fluo-rescence quantitative PCR showed that the FABP3 gene was widely expressed in the tissues of Huang-huai sheep,with the highest relative expression in kidney and the lowest relative expression in rumen.There was no significant difference in the expression of FABP3 in the longissimus dorsi of 3 months old,9 months old,and 18 months old Huanghuai sheep.The expression level of FABP3 gene was sig-nificantly positively correlated with arachidonic acid,trideconic acid and muscle tenderness,but not significantly correlated with IMF and others indicators.[Conclusion]These results indicated that FABP3 gene plays an important role in regulating the meat quality of Huanghuai sheep,and can be used as an important candidate gene for IMF deposition and meat quality regulation in Huanghuai sheep.关键词
