Effects of conjugated linoleic acid on the milk fat globules size and ruminal microbiome of dairy goats

Abstract
[Objective]The aim of this study was to evaluate the effects of conjugated linoleic acid(CLA)on the size and distribution of MFG and ruminal microbiome during milk fat depression in dairy goats,and furthur explore the mechanism of reduced milk fat content.[Method]Twenty-four mid-lactation Saanen dairy goats were randomly divided into four groups with six goats per group.The control group(CON)was fed the basal diet every day,while the experimental groups were divided into low(L-CLA),medium(M-CLA)and high(H-CLA)dose groups,which were supplemented with 30,60 and 90 g·d-1 CLA per group on the basic diet,respectively.The experiment lasted for 21 d after 5 d adaptation period.Natural goat milk was stained with Nile Red and fluorescein isothiocyanate(FITC),and the mean particle size and size distribution of MFG in the four groups was measured.And on the 21st day,ruminal fluids were collected for 16S rRNA gene sequencing to analyze their microbiome.[Result]The results showed that feeding CLA significantly decreased milk fat content of the milk composition.Compared with the control group,the particle size of MFG in the CLA-fed dairy goats was significantly decreased,with a significant dose-dependent effect.In addition,after feeding CLA,the proportion of large MFG decreased and the proportion of small MFG increased.The results of 16S rRNA gene sequencing showed that CLA-induced milk fat depression(MFD)in dairy goats showed a significant decrease with the change of ruminal bacteria abundance,and the relative ruminal bacteria abundance showed significant changes,most of which belonged to the Firmicutes and Bacte-roidetes.The relative abundance of Rikenellaceae_RC9_gut_group,norank_f__Bacteroidales_RF16_group(belonging to Bacteroidetes)and UCG-002(belonging to Firmicutes)in CON group was signifi-cantly higher than that in the H-CLA group.While,the relative abundance of Lachnospiraceae_NK3A20_group,Christensenellaceae_R-7_group,norank_f_UCG-011,norank_f_norank_o_Clos-tridia_UCG-014,norank_f_Eubacteriumicostanioligenes_group,and unclassified_f__Lachnospira-ceae in Firmicutes and norank_f_Maurbaculaceae in Bacteroidetes was significantly lower in the H-CLA group.[Conclusion]In conclusion,CLA altered the distribution of MFG size proportion,thereby reducing the average size of MFG,leading to a decrease in milk fat content,and changing the diver-sity and composition of ruminal microbiome.关键词
