摘要

Abstract

Objective:To investigate the mechanism of Jinshui Huanxian Formula combined with acupuncture in the treatment of pulmo-nary fibrosis(PF)in rats with epithelial-mesenchymal transition(EMT)and transforming growth factor-β1(TGF-β1)/Smad2/3 pathway.Methods:A total of 56 SD rats were randomly enrolled in blank group(group C),model group(group M),sham acupuncture group(group P),acupuncture group(group A),Jinshui Huanxian Formula group(J group,7.83 g·kg-1),combined group(AJ group)and pirfenidone group(group B,100 mg·kg-1),with 8 rats in each group.Except for group C,the other rats were prepared by tracheal instillation of bleomycin.After 29 days of intervention,the lung function indexes of rats were measured,including inspiratory ca-pacity(IC),forced vital capacity(FVC),functional residual capacity(FRC)and dynamic lung compliance(Cydn).HE staining and Masson staining were used to detect the pathological morphology of rat lung,and alveolitis score and PF score were obtained.Immuno-histochemistry was used to detect the protein expression level of α α-smooth muscle actin(α-SMA)in rat lungs.The mRNA expres-sion levels of E-cad,N-cadherin(N-cad)and Vim in lung were detected by RT-qPCR.Western blot was used to detect the pro-tein expression levels of N-cad,p-Smad2,p-Smad3 and TGF-β1 in lungs.Results:Compared with group C,the levels of IC,FVC,Cdyn and FRC in group M decreased(P<0.01).Compared with group M,the levels of IC,FVC(except group A),Cdyn and FRC(except group J)in the four intervention groups were increased(P<0.05).Compared with group A,the levels of IC,FVC,Cdyn and FRC in AJ group were increased.Compared with group C,the alveolitis score and PF score of group M were increased(P<0.01).Compared with group M,the alveolitis score(except group A)and PF score of the four intervention groups were reduced(P<0.05).Compared with group B,group J and group A,the PF score of the rats in the AJ group was decreased(P<0.01).Compared with group C,the expression level of α-SMA protein and the content of HYP in the lungs of group M were increased(P<0.01).Compared with the M group and the P group,the expression level of α-SMA protein and the content of HYP in the lungs of the four intervention groups decreased(P<0.01).Compared with group B,group J and group A,the expression level of α-SMA protein in the lungs of rats in AJ group decreased(P<0.01).Compared with group C,the levels of E-cad mRNA in the lungs of group M decreased,and the levels of N-cad mRNA and Vim mRNA increased(P<0.01).Compared with group M,the levels of E-cad mRNA in the lungs of the four intervention groups increased,and the levels of N-cad mRNA and Vim mRNA decreased(P<0.01).Compared with group B,group J and group A,the levels of E-cad mRNA in AJ group increased,and the levels of N-cad mRNA and Vim mRNA decreased(P<0.01).Compared with group C,the protein expression levels of p-Smad2,p-Smad3 and TGF-β1 in the lungs of group M were increased(P<0.01).Compared with group M,the protein expression levels of p-Smad2(except group A),p-Smad3 and TGF-β1 in the lungs of the four intervention groups were decreased(P<0.01).Conclusion:Jinshui Huanxian Formula combined with acupunc-ture can improve lung function and regulate EMT in rats with pulmonary fibrosis,and the mechanism may be related to the inhibition of TGF-β1/Smad2/3 pathway.
