从氧化应激探讨肌萎缩侧索硬化症的发病机制及相关天然药物的筛选
Pathogenesis of Amyotrophic Lateral Sclerosis from Oxidative Stress and Screening of Related Natural Medicines

Abstract
Objective:To investigate the pathogenesis of amyotrophic lateral sclerosis(ALS)from the perspective of oxidative stress through bioinformatics,and to screen natural drugs that may intervene in ALS,to provide new ideas for the treatment of ALS.Methods:The datasets related to ALS and meeting the screening conditions were retrieved in the GEO database,and DESeq2 differential analysis was performed to obtain the differential genes of ALS.Genes associated with oxidative stress were collected in the GeneCards database.A protein-protein interaction(PPI)network was constructed to common genes for differential expression of ALS and oxidative stress.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of ALS targets related to oxidative stress were performed by R software.The CMap platform was used to screen the natural active ingredients that intervened in ALS,and TCMSP was used to find the source of the natural active ingredients.Results:A total of 1 224 differential genes were obtained from the GSE124439 dataset in the GEO database.A total of 13 632 oxidative stress-related targets were collected and intersected with ALS dif-ferential genes,and imported into the STRING database to obtain the PPI network between ALS targets related to oxidative stress,and 10 core targets were screened.Enrichment analysis showed that the common targets of ALS and oxidative stress were related to inflamma-tion,cell proliferation and chemotaxis.The phospholipinositide 3-kinase/protein kinase B signaling pathway and the interaction be-tween cytokines and their specific receptors ranked high in KEGG analysis.The screening results of CMap platform showed that celand-erythrine,norporine hairline,quercetin marigold,honokiol,umbelliferol,hypericin and papaverine were the most likely natural active in-gredients for the treatment of ALS related to oxidative stress,and these active ingredients were widely derived from a variety of tradition-al Chinese medicines,and the molecular docking results showed that the binding energy of natural active ingredients and their corre-sponding targets was less than-5.0 kcal/mol.Conclusion:Oxidative stress and neuroinflammation are inseparable from the pathogene-sis of ALS,and the seven natural active ingredients screened in this article may have potential therapeutic effects on ALS with oxidative damage.关键词
