摘要

Abstract

Background:Modulation of the dopaminergic neurotransmission system,neuroinflammation,and neurotrophic factors is believed to be beneficial for Attention-Deficit/Hyperactivity Disorder(ADHD)treatment.Tetrahydroxy stilbene glucoside(TSG)has neuroprotective effects,but it remains unclear whether these effects are related to the aforementioned processes. Objective:This study aimed to investigate the effect of TSG on alleviating ADHD-related symptoms by modulating neuroinflammation and the brain-derived neurotrophic factor(BDNF)signaling pathway through bioinformatic analysis and experimental verification. Methods:A network-pharmacology approach was employed to explore the potential targets of tetrahydroxy stilbene glucoside(TSG)on ADHD.Then,the effects of TSG on ADHD related symptoms were examined by animal experiment,the neuroinflammation and BDNF-related signaling pathways were also examined.Wistar Kyoto(WKY)rats were used as normal animals.Junior spontaneously hypertensive rats(SHRs)were assigned to the model group,positive control and TSG(low,medium,and high dose)groups.Behavioral alterations were assessed using the open field test(OFT)and Morris water maze(MWM).Pro-inflammatory cytokines and BDNF-related proteins in prefrontal cortex,striatal,and hippocampal samples were analyzed via immunochemical staining,qRT-PCR,and immunoblot. Results:Compound-target-pathway data indicated that TSG may improve ADHD via the key targets such as AKT1,BDNF,SOS1,etc.TSG alleviated hyperactivity and impulsivity in SHRs and improved spatial memory.TSG decreased docosahexaenoic acid(DHA)levels while elevating Akt1,Bdnf,Sos1,Pik3cg,Gab1,and Ntrk2 mRNA levels.Protein levels of SOS1,GAB1,AKT1,TrkB,and kinase P110 beta were also elevated in the prefrontal cortex,striatal,and hippocampal samples. Conclusion:TSG improves ADHD symptoms in SHR rats,partially through the inhibition of neuroinflammation and the regulation of neurotrophic factors.
