摘要

Abstract

[Objectives]This paper aimed to identify the target genes regulated by the cassava TGA transcription factor MeTGA9.3,and lay the foundation for further studying the function of this transcription factor in regulating the synergistic and efficient utilization of water and nitrogen nutrients in cassava.[Methods]The transcription factor characteristics of MeTGA9.3 were determined based on subcellular localization and transcriptional activation experiments.DAP-Seq was used to identify the DNA elements recognized and bound by MeTGA9.3 at the whole genome level.[Results]The MeTGA9.3 protein localized in the nucleus and exhibited significant transcriptional activation activity in yeast.The MeTGA9.3 promoter drove the reporter gene GUS significantly expressing in the roots of transgenic Arabidopsis thaliana.The advantageous DNA element for MeTGA9.3 transcription factor recognition and binding was TGACGTCA.In the predicted target genes of MeTGA9.3,several LEA,ERF,and MYC members related to stress were found,as well as three members of nitrogen transport protein 1(NRT1).[Conclusions]This study characterized cassava TGA transcription factor MeTGA9.3,and found that MeTGA9.3 had the potential to synergistically regulate drought stress response and nitrogen transport.
