Effects of different light qualities and rootstocks on photosynthetic characteristics and sugar and acid metabolism in'Kyoho'grape leaves

Abstract
[Objectives]The purpose of this paper was to study the effects of different light qualities and rootstocks on the photosynthetic characteristics and sugar and acid metabolism in the leaves of'Kyoho'grape plants.[Methods]The'Kyoho'grape selfrooted seedlings and grafted seedlings with'3309C''1103P'and'Kangzhen 3'as rootstocks were used as test materials.LED(blue light,red light,green light and white light)was used for illumination.The growth status and leaf traits of the plants under different treatments were investigated,and the photosynthetic indices,chlorophyll fluorescence parameters,soluble sugar and organic acid components were determined.The expression levels of genes related to photosynthesis and sugar metabolism pathways were also determined.[Results]Blue light treatment increased the chlorophyll content of the four stock-scion combinations,and the chlorophyll contents of the combinations of'Kyoho'/'3309C','Kyoho'/'1103P',and'Kyoho'/'Kangzhen3'were higher than those of the self-rooted seedlings.Red light treatment had a significant promoting effect on the growth and development of plants.The plant height and leaf area of the combination of'Kyoho'/'Kangzhen 3'were higher than those of other combinations.Blue light treatment increased the net photosynthetic rate(Pn)of leaves,while green light treatment inhibited the net photosynthetic rate;the net photosynthetic rate of the combination of'Kyoho'/'3309C'was the highest.Green light treatment increased the Tr of leaves,and the transpiration rates of the two combinations of'Kyoho'/'1103P'and'Kyoho'/'Kangzhen3'were the highest.Light treatment had a greater effect on glucose content.The sugar contents of the combinations of'Kyoho'/'3309C'and'Kyoho'/'1103P'were the highest,and the sugar content of the self-rooted seedlings of'Kyoho'was the lowest.Different light quality treatments affected the content of organic acids,among which red light and blue light had the greatest impact on the content of organic acids.Red light and blue light treatments significantly upregulated the expression levels of photosynthetic pathway-related genes VvLhc,VvPsbS,VvPsbQ and sugar metabolism pathway-related genes VvCWINV,VvHXK2,and VvFRK.[Conclusions]Rootstocks significantly enhanced light responsiveness in'Kyoho'grapevines.The rootstock combinations'Kyoho'/'3309C'and'Kyoho'/'1103P'demonstrated superior sensitivity to red light and blue light spectra,providing a theoretical basis for optimizing light environment regulation in facility-based grape cultivation.关键词
