Abstract

[Objectives]The paper aimed to identify the main pathogenic fungi that cause chrysanthemum stem rot disease.[Methods]The chrysanthemum with decaying stem was collected from the chrysanthemum planting base as the research materials,and the tissue at the junction of diseased and healthy areas was collected and the pathogenic fungi were isolated and purified using the tissue separation method.Subsequently,the rDNA-ITS and EF-1α sequences in the pathogenic fungi were cloned through polymerase chain reaction and sequenced,and a phylogenetic tree was constructed based on these sequences to conduct molecular biological identification of these isolated fungi.The types of pathogens were further confirmed by observing the morphology of hyphae and conidia.The pathogenicity and the Koch postulates assays confirmed that the isolated strains were the pathogenic fungi that caused the rot disease of chrysanthemum stem.[Results]A total of 10 diseased plants with rot stems were collected in this study,and 19 strains of pathogenic fungi were isolated and identified,including 9 strains of Fusarium solani,8 strains of Fusarium commune,and 2 strains of Fusarium oxysporum,all of which belonged to Fusarium species.The pathogenicity assays showed that the isolated strains were all pathogenic to chrysanthemum,and the strains showed different levels of pathogenicity to chrysanthemum.Among the pathogenic fungi,FN3-3 was the most pathogenic strain of F.solani,DT3-3 was the most pathogenic strain of F.oxysporum,and HY1-3 was the most virulent of F.commune.[Conclusions]This study clarified the main pathogenic fungal types of chrysanthemum stem rot disease and provided important clues and basis for the formulation of effective control methods for stem rot disease,the screening of stem rot resistant varieties and disease-resistant breeding.
