慢性冷刺激条件下Ogt基因缺失对小鼠肝脏线粒体稳态的影响
Impact of Ogt Gene Deletion on Mitochondrial Homeostasis in the Liver of Mice under Chronic Cold Stimulation

Abstract
To investigate the role of O-linked β-N-acetylglucosamine modification(O-GlcNAc)glycosylation in regulating mitochondrial homeostasis under chronic cold stimulation conditions,we utilized wild-type(WT)and liver-specific knockout Ogt(Ogt lKO)C57BL/6 mice as experimental animals.These mice were randomly assigned to four groups:WT control group,WT cold stimulation group,Ogt lKO control group,and Ogt lKO cold stimulation group.Western blot was employed to assess the levels of O-GlcNAcylation and OGT expression in the liver tissues of WT and Ogt lKO mice before and after chronic cold stimulation.Additionally,the expression levels of Dynamin-related protein 1(DRP1),Mitofusin-1(MFN1),PTEN induced putative kinase 1(PINK1),Parkin protein(Parkin),and Per-oxisome proliferator-activated receptor γ coactivator 1-alpha(PGC-1α)in mitochondria were also evaluated.The findings revealed that chronic cold stimulation significantly elevated the expression levels of OGT and O-GlcNAcylation in the liver of WT mice(P<0.05),as well as the expression levels of DRP1 and MFN1 in mitochon-dria(P<0.05).However,there were no significant differences in the expression levels of PINK1,Parkin,and PGC-1α among these groups.Compared to WT mice,Ogt lKO mice exhibited significantly elevated expression levels of DRP1,MFN1,and PINK1 in their mitochondria after chronic cold stimulation(P<0.05),while there were no signifi-cant differences in the expression levels of Parkin and PGC-1α.These results suggest that under chronic cold stimu-lation conditions,mitochondrial functional homeostasis becomes imbalanced in mouse liver tissue,leading to mito-chondrial autophagy.Furthermore,Ogt deficiency can accelerate this process.关键词
