Cloning,identification of chemokine gene ccl8 in grass carp(Ctenopharyngodon idella)and its expression responses to Aeromonas hydrophila and Ichthyophthirius multifiliis infections

Abstract
To enrich the related research on the immune regulatory mechanism of chemokines in grass carp(Ctenopharyngodon idella),the ccl8 gene was cloned.The full-length of ccl8 cDNA is 675 bp,the open reading frame is 315 bp,and it encodes 104 amino acids.This gene displays typical characteristics of the chemokine superfamily,with four conserved cysteine residues at amino acids positions 31,32,55,and 71.Amino acid se-quence homology analysis indicated that the ccl8 gene of grass carp had a relatively high homology(88.35%)with the ccl8-like gene of Megalobrama amblycephala.Real-time fluorescence quantitative PCR showed that the ccl8 mRNA was expressed in 10 different tissues of healthy grass carp.The expression level was highest in the immune tissues hepatopancreas and spleen,and relatively high in the head kidney,skin,intestine,gills and heart.How-ever,the expression level was the lowest in the blood,brain and muscle.This indicates that the ccl8 gene is generally expressed in different tissues of grass carp.After being infected by Aeromonas hydrophila,the expression of ccl8 mRNA in the hepatopancreas rapidly increased at 6 hours post-infection.In contrast,the peak expression levels in the head kidney and spleen were observed at 9 hours and 12 hours post-infection,respectively.The expression levels in tissues such as skin and gills didn't change significantly.After being infected by Ichthyoph-thirius multifiliis,the expression of ccl8 mRNA in the skin and spleen tissues reached its peak at 48 hours post-infection,while the peak expression in the gills and head kidney was observed at 36 hours post-infection.The results indicated that the ccl8 genes can promptly respond to infections by Aeromonas hydrophila and Ichthyoph-thirius multifiliis in different tissues,with responses related to the invasion sites.This prompt response potentially contributes to immune regulation in grass carp and plays a critical role in defending against pathogen infections.关键词
