Correlation Between Serum Levels of miR-31-5p,miR-379-5p,and miR-142-3p with Disease Activity and Renal Inju-ry in Patients with Systemic Lupus Erythematosus

Abstract
Objective To investigate correlations among serum microribonucleic acid-31-5p(miR-31-5p),miR-379-5p,and miR-142-3p with disease activity and renal injury in patients with systemic lupus erythematosus(SLE).Methods From Jan-uary 2022 to April 2023,102 SLE patients admitted to our hospital were regarded as SLE group.According to the KDIGO clinical practice guidelines for acute renal injury,29 SLE patients with renal injury were as the renal injury group,while 73 SLE patients without renal injury were as the non-renal injury group.Total 102 healthy volunteers who underwent physical examinations in our hospital were included as control group.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)was applied to detect miR-31-5p,miR-379-5p,and miR-142-3p in serum.ROC curve was applied to analyze the diagnostic value of serum miR-31-5p,miR-379-5p,and miR-142-3p for renal injury in SLE patients.The relationship between serum miR-31-5p,miR-379-5p,and miR-142-3p with disease activity and renal injury was analyzed using Spearman correlation analysis.Results The miR-31-5p the SLE group was obviously reduced compared to control group,while miR-379-5p and miR-142-3p were obviously in-creased(P＜0.05).The miR-31-5p in the serum of SLE patients showed severe activity group＜moderate activity group＜mild ac-tivity group＜no activity group,miR-379-5p and miR-142-3p showed a trend of severe activity group＞moderate activity group＞mild activity group＞no activity group(P＜0.05).The serum miR-31-5p in the renal injury group was obviously lower than that in non-renal injury group,while miR-379-5p and miR-142-3p were obviously increased(P＜0.05).The AUC of the combination of serum miR-31-5p,miR-379-5p,and miR-142-3p in diagnosing renal injury in SLE patients was obviously higher than that of single diagnosis(Z=2.965,P=0.003,Z=3.913,P＜0.001,Z=3.930,P＜0.001).Spearman correlation showed that serum miR-31-5p in SLE patients was negatively correlated with renal injury and disease activity(rs=-0.541,P＜0.001,r,=-0.523,P＜0.001),while miR-379-5p and miR-142-3p levels were positively correlated with renal injury(r,=0.481,P＜0.001,rs=0.503,P＜0.001)and disease activity(rs=0.498,P＜0.001,r,=0.539,P＜0.001).Conclusion The serum miR-31-5p level in SLE patients is obviously reduced,while miR-379-5p and miR-142-3p levels are obviously increased,which are related to disease activity and renal injury.关键词
