白藜芦醇调节TREM-1表达减轻大鼠脑缺血再灌注损伤后细胞焦亡
Resveratrol Regulates TREM-1 Expression to Alleviate Cell Pyroptosis After Cerebral Ischemia-Reperfusion Injury in Rats

Abstract
Objective To investigate the mechanism of resveratrol in alleviating cell pyroptosis following a cerebral ische-mia-reperfusion injury in rats.Methods The adult Sprague-Dawley(SD)rats were chosen to establish a middle cerebral artery occlusion/reperfusion(MCAO/R)injury model.Rats were randomly divided into the following groups:Two groups:Sham and MCAO/R,Four groups:Sham,MCAO/R,MCAO/R+Res(Resveratrol),and MCAO/R+Vehicle,Five groups:Sham,MCAO/R,MCAO/R+Res,MCAO/R+Res+PBS,and MCAO/R+Res+PY159.Neurological deficits were assessed using the modified Be-derson score and modified Neurological Severity Score(mNSS).The expression of TREM-1 and NLRP3 in ischemic brain tissue was detected by immunofluorescence.qPCR was performed to measure the mRNA levels of TREM-1,IL-1β,and NLRP3.Western blot(WB)was used to analyze protein expression of TREM-1,NLRP3,and cleaved Caspase-1(Cl-Caspase1).The levels of IL-1βand IL-18 were quantified by ELISA.Results After MCAO/R treatment,the qPCR,immunofluorescence,WB,and ELISA tests data showed increased expression levels of TREM-1,NLPR3,Cl-Caspase1,IL-1β and IL-18.Compared to the MCAO/R group,the expression levels of TREM-1,NLPR3,Cl-Caspase1,IL-1β and IL-18 were significantly decreased in the MCAO/R+Res group(P<0.05),along with a reduction in the modified mNSS score and modified Bederson score.Nevertheless,PY159 was able to counteract the inhibitory effects of resveratrol on cell apoptosis and neuroprotective effects following MCAO/R(P<0.05).Con-clusion Resveratrol may reduce cell apoptosis after cerebral ischemia-reperfusion by decreasing the expression of TREM-1,there-by exerting neuroprotective effects.关键词
