A Microfluidic Chip for Drug Screening in Head and Neck Tumors

Abstract
Objective:This study designed an automatic microfluidic chip for drug screening of head and neck tumor cells to improve the throughput and rapidity.Methods:Firstly,the design includes minimal chamber volumes for head and neck squamous cell carcinoma(HNSCC)organoid culture,and dual-channel concentration gradient generator.Then,the stability of the chip was verified through simulation and actual operation.Finally,we compared the cell viability inside and outside the chip after drug screening.Results:The concentration gradient based on convection generates six different drug concentra-tions,allowing for stable drug mixing within 100 μL/min.Cell viability of HNSCC was able to decrease with increasing drug concentration during the test,and the results inside and outside the chip were consistent.Conclusion:The study validates the feasibility of microfluidic chip in drug screening on head and neck tumor cells,providing a significant opportunity for the development of personalized treatment plans for head and neck tumors.关键词
