丹皮酚对脑胶质瘤细胞增殖和侵袭能力的影响及机制探讨
Effects of paeonol on proliferation and invasion of glioblastoma cells and their underlying mechanisms

Abstract
Objective To investigate the effects of paeonol on the proliferation and invasion of human glioblastoma cells and their regulatory mechanisms on the phosphatidylinositol 3-kinase/protein kinase B/extracellular signal-regulated kinase(PI3K/AKT/ERK)signaling pathway.Methods Human glioblastoma cell line GL15 in the logarithmic growth phase was divided into four groups:control group(with culture medium only,no drug treatment),insulin-like growth factor-1(IGF-1)group(treated with 100 ng/mL IGF-1,an activator of PI3K/AKT/ERK signaling pathway),paeonol group(treated with 200 mg/L paeonol),and paeonol+IGF-1 group(co-treated with 200 mg/L paeonol and 100 ng/mL IGF-1).Cell proliferation rates were measured by CCK-8 assay,cell invasion was assessed by Transwell assay,and the protein expression levels of phosphorylated phosphatidylinositol 3-kinase(p-PI3K),phosphorylated protein kinase B(p-AKT),and phosphorylated extracellular signal-regulated kinase(p-ERK)in GL15 cells were detected by Western blot-ting.Results Compared with the control group,the IGF-1 group showed increased proliferation rates at 24 and 48 h(both P<0.05).The paeonol group exhibited decreased proliferation rates as compared with the other three groups at 24 and 48 h(all P<0.05).Compared with the control group,the paeonol group had reduced number of invasion cells while the IGF-1 group showed increased invasion cells(all P<0.05).Compared with the IGF-1 group,the paeonol group demon-strated decreased invasion cells(P<0.05).Compared with the control group,the protein expression levels of p-PI3K,p-AKT,and p-ERK decreased in the paeonol group,while the protein expression levels of p-PI3K,p-AKT,and p-ERK in-creased in the IGF-1 group(all P<0.05).Compared with the paeonol+IGF-1 group and IGF-1 group,the expression lev-els of p-PI3K,p-AKT,and p-ERK significantly decreased(all P<0.05).Conclusion Paeonol can inhibit the prolifera-tion and invasion of GL15 cells,and thein mechanism may be related to the inhibition of activation of PI3K/AKT/ERK sig-naling pathway.关键词
