摘要

Abstract

Objective To explore the effect of the Mongolian medicine Eridun Wurile on the antioxidant indexes,inflammatory factors,and nuclear feator-e2-related factor 2/heme oxygenase 1(Nrf 2/HO-1)signaling in brain tissues of rats with ischemic vertigo.Methods Eighty-four SD rats were randomly assigned into a sham surgery group(Sham),a model group(M),Eridun Wurile groups high dose(EW-H,1.08 g/kg),middle dose(EW-M,0.54 g/kg),and low dose(EW-L,0.27 g/kg),as well as a Jinnardo group(JND,60 mg/kg)using a random number table,with 14 rats in each group.The Sham group received the right common carotid artery(CCA)and subclavian artery(SCA)dissection,without ves-sel ligation.All the other groups received CCA and SCA ligation to establish a rat model of ischemic vertigo.After success-ful modeling,medicines were continuously administered for 7 days according to the different medicines of each group.After-ward,the following aspects were observed in each group:brain tissue pathological changes by hematoxylin-eosin(HE)staining,tumor necrosis factor-a(TNF-α),interleukin-1 β(IL-1β),superoxide dismutase(SOD),and malondialde-hyde(MDA)protein content by enzyme-linked immunosorbent assay(ELISA),and protein expression of Nrf 2 and HO-1 pathway in brain tissue by immunohistochemical staining.Results Compared with the Sham group,the nerve cells in the brain tissue of group M rats were severely damaged,the TNF-α,IL-1 β,and MDA content were significantly increased(P<0.05),while the SOD content and protein expression of Nrf 2 and HO-1 were significantly decreased(P<0.05).Compared with the M group,the nerve cells in the brain tissue were improved in the other groups.Specifically,the TNF-α,IL-1 β,and MDA content were decreased,while the positive expression of SOD,as well as Nrf 2 and HO1 protein was in-creased.Specifically,the EW-H group and the JND group(P>0.05),the EW-H and the JND groups with the EW-M and the EW-L groups(P<0.05),as well as the EW-M group and the EW-L group(P<0.05)were compared.Con-clusion Mongolian medicine Eridun Wurile may exert its protective effect on ischemic vertigo by enhancing antioxidant capacity,inhibiting inflammatory factor secretion,and initiating the Nrf 2/HO-1 pathway.Besides,there is a dose-effect relationship.
