中药复方治疗糖皮质激素性骨质疏松的配伍规律聚类分析及作用机制的网络药理学研究
Cluster Analysis of Compatibility Patterns and Network Pharmacology Study on Mechanisms of Traditional Chinese Medicine Compound Formulas in Treating Glu-cocorticoid-induced Osteoporosis

Abstract
Objective To investigate the current use of traditional Chinese medicine(TCM)compound formulas in the treatment of glucocorticoid-induced osteoporosis(GIOP),clarify the compatibility patterns through data mining,and explore the potential therapeutic targets and mechanisms of action of high-frequency herbal drugs used in these formulas.Methods Literature on TCM compound formulas for GIOP published from database inception to May 21,2024,was re-trieved from CNKI,VIP,and Wanfang Data.Data were collected and analyzed using IBM SPSS Statistics 24 and Microsoft Excel 2022.Clustering analysis and association rule mining were performed using SPSS Modeler 18.0.The chemical com-ponents and targets of the high-frequency herbal drugs were obtained from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Potential disease targets were identified by searching and screen-ing the GeneCards and Online Mendelian Inheritance in Man(OMIM)databases.Core targets were determined by intersec-ting drug and disease targets,and visualized with a Venn diagram.These core targets were imported into the STRING database to construct a protein-protein interaction(PPI)network,from which final key targets were identified.Functional enrich-ment analysis(GO)and pathway analysis(KEGG)were conducted using the David platform.Results A total of 29 TCM compound formulas comprising 90 unique herbal drugs with a cumulative usage frequency of 320 were included.The top five core herbal drugs were Rehmanniae Radix Praeparata,Dioscoreae Rhizoma,Epimedii Folium,Eucommiae Cortex,and Achyranthis Bidentatae Radix.These five herbal drugs yielded 65 active compounds and 227 corresponding targets.A total of 707 potential disease-related targets were identified,with 74 overlapping targets considered as core drug-disease tar-gets.GO enrichment analysis revealed 485 biological processes,39 cellular components,and 73 molecular functions,while KEGG analysis identified 168 signaling pathways,primarily involving cell activation,cytokine receptor binding,positive reg-ulation of intra-and extracellular processes,and inflammatory responses.Key pathways included the MAPK signaling pathway,IL-17 signaling pathway,and HIF-1 signaling pathway.Conclusion TCM compound formulas for treating GIOP primarily focus on nourishing the liver and kidneys and replenishing essence and marrow,supplemented by tonifying qi and blood and promoting blood circulation to remove stasis.The core herbal drugs exert their therapeutic effects through multiple components,targets,and pathways,closely associated with targets such as AKT1 and TNF,and act via the MAPK,IL-17,and PI3K-Akt signaling pathways.These findings provide valuable reference for future research and clinical treat-ment of GIOP.关键词
