Abstract

Objective:To explore the mechanism by which Yupingfeng combined with Erchen Powder alleviates inflammatory re-sponses in a rat model of cough variant asthma(CVA)by targeting NOD-like receptor protein 3(NLRP3),apoptosis-associated speck-like protein(ASC),and caspase-1.Methods:Healthy male rats were divided into control,model,dexamethasone,and Yup-ingfeng+Erchen Powder group.A CVA model was established using ovalbumin(OVA)and aluminum hydroxide sensitization.Pul-monary inflammation was assessed by hematoxylin-eosin(HE)staining.Serum levels of interleukin(IL-1β)and IL-18 were meas-ured using enzyme-linked immunosorbent assay(ELISA).The expression of NLRP3,ASC,and caspase-1 at both mRNA and protein levels in lung tissues was evaluated by real-time quantitative PCR and Western blot.Results:Yupingfeng combined with Erchen Powder alleviated lung tissue inflammation in CVA rats,significantly reduced serum levels of the inflammatory cytokines IL-1β and IL-18,and downregulated the mRNA and protein expression of NLRP3,ASC,and caspase-1 in lung tissues.Conclusion:Yuping-feng combined with Erchen Powder may exert therapeutic effects on CVA by attenuating inflammatory responses through the down-regulation of NLRP3,ASC,and caspase-1 expression.
