糖肾宁抑制系膜增殖改善糖尿病肾病肾纤维化的作用机制
Mechanism of Tangshenning in Inhibiting the Proliferation of Mesangial Cells to Improve Renal Fibrosis in Diabetic Kidney Disease

Abstract
Objective:To investigate the mechanism by which Tangshenning(TSN)inhibits mesangial cell proliferation and amel-iorates renal fibrosis in diabetic kidney disease(DKD)based on the Yes-associated protein(YAP)/PDZ-binding motif transcrip-tional coactivator(TAZ)signaling pathway.Methods:A DKD mouse model was established by feeding KK-Ay mice a high-fat diet.Successfully modeled mice were randomly divided into three groups:model group,TSN group,and valsartan group,with C57BL/6J mice used as the normal control.After 10 weeks of continuous administration,body weight,blood glucose,blood urea nitrogen(BUN),serum creatinine(Scr),and urinary albumin-to-creatinine ratio(UAER)were measured.Renal histopathology was evalua-ted by hematoxylin-eosin(HE),periodic acid-Schiff(PAS),and Masson's trichrome(Masson)staining.Ultrastructural changes in glomeruli were observed by transmission electron microscopy.Protein expression levels of YAP,TAZ,TEA domain transcription fac-tor 1(TEAD1),connective tissue growth factor(CTGF),and type Ⅳ collagen(Col-Ⅳ)in renal tissues were detected using West-ern blot.Expression of fibronectin(FN)and laminin(LN)was assessed by immunohistochemistry.Results:Compared with the model group,TSN had no significant effect on body weight and blood glucose,but significantly reduced BUN and Scr levels.UAER also decreased significantly after treatment(P<0.05).Protein expression levels of YAP,TAZ,TEAD1,CTGF,and Col-Ⅳ in renal tissue were significantly downregulated(P<0.05).FN and LN expression levels were also reduced.Histological analysis revealed alleviation of mesangial matrix,mesangial cell proliferation,glycogen and collagen deposition,as well as reduction in glomerular basement membrane thickening,foot process fusion,and extracellular matrix accumulation in the mesangial area.Conclusion:TSN may inhibit mesangial cell proliferation and extracellular matrix accumulation by suppressing the YAP/TAZ signaling pathway,thereby improving renal fibrosis in DKD.关键词
