荆防颗粒改善大鼠寒湿型变应性鼻炎的实验研究
Experimental Study of Jingfang Granules in Ameliorating Cold-dampness Type Allergic Rhinitis in Rats

Abstract
Objective:To establish a cold-dampness type allergic rhinitis(AR)rat model and evaluate the therapeutic effects and mechanism of Jingfang Granules on cold-dampness type AR.Methods:Sprague Dawley(SD)rats were randomly divided into con-trol,model,loratadine(positive control),and low-,medium-,and high-dose Jingfang Granules treatment groups.The cold-dampness type AR model was established by sensitizing the rats with ovalbumin(OVA)combined with exposure to a cold and humid environ-ment.After the final intranasal OVA challenge,nasal symptoms in each group were recorded and scored.The spleen index was cal-culated,and serum levels of OVA-specific IgE(OVA-sIgE),interferon-γ(IFN-γ),interleukin-4(IL-4),IL-5,and IL-13 were meas-ured using enzyme-linked immunosorbent assay(ELISA).Nasal mucosal histopathological changes were observed via hematoxylin and eosin(HE)staining.The mRNA expression levels of PTEN-induced kinase 1(PINK1),E3 ubiquitin ligase Parkin,microtu-bule-associated protein 1 light chain 3(LC3),sequestosome 1(p62),and translocase of the outer mitochondrial membrane 20(TOM20)in nasal tissues were determined using quantitative real-time PCR(qRT-PCR).Results:Compared with the control group,the model group exhibited pronounced AR symptoms,elevated spleen index,significant nasal mucosal damage,and increased levels of OVA-sIgE,IL-4,IL-5,and IL-13.Compared with the model group,rats treated with Jingfang Granules showed significantly reduced symptom scores,decreased inflammatory cell infiltration around nasal epithelial cells(as shown by HE staining),and im-proved inflammatory mediator levels in serum.Moreover,the mRNA expression of PINK1,Parkin,and LC3 was upregulated,while the expression of p62 and TOM20 was downregulated in nasal mucosa tissue.Conclusion:Jingfang Granules can alleviate symptoms of cold-dampness type AR in rats,potentially by regulating the PINK1/Parkin signaling pathway and enhancing mitophagy.关键词
