Abstract

[Aim]Isolation and identification of Pseudomonas syringae pv.pisi type Ⅰ on imported pea seeds from United States.[Method]An isolate 5337-2 was obtained on pea seeds imported from United States with semi-selective MT medium,and detected by the pathogenicity,hypersensitive reaction,PCR detection,16S rDNA sequence analysis,multilocus sequence typing and bio-chemical tests.[Result]The isolate could cause the top young stalk to wither when inoculated artificially with pea stalks.The isolate could produce anaphylactic reaction when inoculated with tobacco leaves.The DNA of isolate 5337-2 could be amplified by the spe-cific primer AN3F/AN3R and probe P-PS1 of Ppi type Ⅰ,producing expected 131bp specific bands and amplification curves.The 16S RNA,16S-23S and partial 23S RNA sequences of isolate 5337-2 were consistent with those strains of Ppi type Ⅰ in GenBank,and the similarity was 100.00%.Four housekeeping genes gap1,gltA,gyrB and ropD were selected for multilocus sequence analy-sis.Phylogenetic tree showed that isolate 5337-2 was in the same branch with Ppi type Ⅰ strains and in different branches with Ppi type Ⅱ strains.The biochemical characteristics of the isolates such as esculin,erythritol and fluorescence were consistent with Ppi type Ⅰ.[Conclusion]Based on these results above,the isolate 5337-2 is identified as Pseudomonas syringae pv.pisi type Ⅰ.
