基于全转录组测序的亚洲柑橘木虱激素生物合成相关基因互作网络构建
Construction of hormone biosynthesis-related gene interaction network in Diaphorina citri based on whole transcriptome sequencing

Abstract
[Aim]Development is a crucial biological activity for increasing the damage caused by Diaphorina citri.This study aims to deepen the understanding of the post-transcriptional regulation mechanism of the D.citri and enrich the molecular information of the developmental regulatory network of the D.citri.[Method]Whole transcriptome sequencing was performed on a mixed sample comprising all developmental stages of D.citri in this study.We mainly focused on the insect hormone biosynthesis signaling pathway to perform new transcript mining,transcriptome variation statistics,gene structure analysis,interactive miRNA prediction,and con-struction of miRNA-LncRNA and miRNA-CircRNA interaction networks.The Pearson bivariate correlation analysis was used to de-termine the correlation between the expressions of candidate miRNAs and hormone-related genes.[Result]In this study,25 new transcripts that could be annotated to the insect hormone biosynthesis signaling pathway were identified.It was found that genes such as JH-o-MT(juvenile hormone acid O-methyltransferase),JHEH2(JH epoxide hydrolase 2),MFE(methyl farnesoate epoxidase),FOHSDR(dehydrogenase/reductase SDR family member),ALDH(aldehyde dehydrogenase),and JHE(JH esterase)may contain previously unannotated untranslated regions,as determined by comparative analysis with existing genomic databases.Through miR-NA prediction,six categories of miRNAs,including miR-252a-5p,miR-315-5p,miR-71-5p,miR-993-3p,miR-1000-5p,and miR-929-5p,were found to interact with FOHSDR and JHEH2,respectively.Further construction of the FOHSDR/JHEH2-miRNA-LncRNA and FOHSDR/JHEH2-miRNA-CircRNA interaction networks revealed eight LncRNAs,including MSTRG.30118.1,MSTRG.16869.1,MSTRG.39461.1,and MSTRG.29283.2,could interact with five categories of miRNAs,while 45 CircRNAs,in-cluding dcicirc 003865,dcicirc 001724,dcicirc 004421,dcicirc 006125,and dcicirc 006016,could interact with six categories of miRNAs.Furthermore,miR-252a-1-5p,miR-315-1-5p,miR-71-2-5p,and miR-929-1-5p exhibited reduced expression levels in response to flush shoots.The qPCR and correlation analysis indicated that the expression of miR-929-5p showed a strong negative correlation with the key JH synthesis rate-limiting enzyme HMGCR.miR-252a-1-5p and miR-929-1-5p also showed a mod-erate negative correlation with the JH synthesis genes FPPS and FA-o-MT and the 20E synthesis gene Sad.[Conclusion]This study is the first report on constructing the interaction networks of FOHSDR/JHEH2-miRNA-LncRNA and FOHSDR/JHEH2-miRNA-Cir-cRNA in D.citri.The study obtained miRNAs,LncRNAs,and CircRNAs with high connectivity.The above results suggest that these miRNAs,LncRNAs,and CircRNAs may play important roles in the post-transcriptional regulation of hormone biosynthesis in the D.citri.It was also found that miR-252a-1-5p and miR-929-1-5p might be significantly associated with initiating JH and 20E bi-osynthesis after D.citri feeding on flush shoots.These results could provide important clues for revealing the post-transcriptional reg-ulation mechanism of the D.citri development.关键词
