Adipokinetic hormone receptor regulates lipid mobilization to mediate ovarian development of Spodoptera litura(Lepidoptera:Noctuidae)

Abstract
[Aim]The adipokinetic hormone receptor(AKHR)plays an important role in insect reproductive regulation processes such as ovarian development,mating,and egg laying.The aim of this study was to investigate the function of the AKHR in the ovari-an development of Spodoptera litura.[Method]In this study,the expression pattern of the AKHR in the adult female of S.litura was analyzed using real-time fluorescence quantitative PCR(qPCR).Gene interference technology(RNA interference,RNAi)was used to reduce the expression of SlAKHR.Ovarian development in female adults was observed,and changes in triglyceride contents in the abdomen of S.litura were measured using physiological and biochemical methods.Furthermore,the expression of genes relat-ed to lipid metabolism and reproduction were analyzed by qPCR.[Result]SlAKHR expression was highest in female fat bodies,be-ing low at the time of first emergence of female adults and then gradually increased,reaching a maximum at 48 h,and then de-creased.After the successful reduction of SlAKHR expression(74%-82%decrease),the number of immature oocytes increased,the size and number of lipid droplets in the ovary became smaller and decreased significantly,and the localization of its vitellogenin in the follicular epithelium was significantly reduced.In addition,significantly suppressed expression of SlAKHR was followed by in-creased accumulation of abdominal triglycerides and significant downregulation in the expression of lipase(brummer),fatty acid synthase,vitellogenin,juvenile-protective hormone receptor(Methoprene-tolerant),and transcription factor Krüppel homolog 1.[Conclusion]The results indicated that the regulation of lipid homeostasis by SlAKHR,mediated through reproduction-related genes,affected the ovarian development ofS.litura.This study provides a scientific basis for the research on green pest control strat-egies based on RNAi.关键词
