Abstract

[Objective]Heavy metal ATPase(HMA)proteins play critical roles in the absorption and transport of various metal ions in plants.This study aimed to characterize the HMA gene family in foxtail millet through genome characterization,expres-sion profiling,and homology analysis to elucidate its functions.[Method]HMA gene family members in foxtail millet were identified using protein homology alignment and conserved domain screening.Bioinformatics analyses were performed to exam-ine their physicochemical properties,gene family expansion,conserved motifs,phylogenetic relationships,and protein molecu-lar docking.Transcriptomic and qRT-PCR analyses under Cd and Fe stress treatments were conducted to assess metal-respon-sive expression patterns.Functional predictions for selected SiHMA were inferred through homology with rice.[Results]Twelve HMA genes were identified and classified into two subgroups.Family expansion involved six HMA genes through two tandem duplication events(four genes,33.3%)and one segmental duplication event(two genes,16.7%).SiHMA showed tis-sue-specific responses to Cd and Fe stress.The possible functions of SiHMA were predicted.Functional predictions suggested that SiHMA5 mediated Zn,Cu,Pb and Cd efflux,while SiHMA10 might be involved in the loading and transportation of Cu in plants.[Conclusion]Tandem and segmental duplication drove HMA family expansion and functional diversification in foxtail millet.Cis-acting elements related to plant hormones and stress responses were identified in SiHMA promoters.Based on ho-mology,expression patterns,and previous evidence,we proposed roles for SiHMA1,SiHMA5,SiHMA6 and SiHMA10 in metal homeostasis.These results provide a theoretical basis for further study on HMA genes in foxtail millet.
