基于BSA-seq的高粱柄伸长度QTL定位
QTL mapping of sorghum peduncle exsertion length using BSA-seq

Abstract
[Objective]This study aimed to identify quantitative trait loci(QTL)associated with sorghum peduncle length using BSA-seq technology,screen sorghum plants with excellent peduncle elongation traits,and provide a theoretical basis for im-proving elite sorghum backbone lines and breeding mechanization-adapted varieties.[Methods]The study used sorghum main-tainer lines 7050B(short peduncle)and Tx2925B(long peduncle),along with their F2 population derived from emasculated hy-bridization.Peduncle length was measured in parents and the segregating population.Two bulked pools,EL-L(long pedun-cle)and EL-S(short peduncle),were constructed for QTL mapping.[Results]The peduncle length of 7050B was significantly shorter than that of Tx2925B,with a phenotypic variation coefficient variation of 76.1%in the F2 population.SNP variations detected in EL-L,EL-S,7050B,and Tx2925B were 1 574 836,2 041 538,2 512 953 and 688 446 respectively.At a 99%confidence level,16 QTLs were identified on chromosomes 1,4,5,8 and 10,including 9 on chromosome 1,4 on chromo-some 4,and 1 each on chromosomes 5,8 and 10.The candidate intervals contained 2 to 1,401 genes,with 2748 and 1779 transcripts annotated in GO and KEGG databases,respectively.[Conclusion]This study identified 16 QTLs significantly asso-ciated with peduncle length across five sorghum chromosomes,providing candidate gene resources for deciphering the genetic mechanisms of peduncle elongation.These findings establish a molecular foundation for breeding mechanized sorghum varieties and improving elite germplasms.关键词
