摘要

Abstract

Objective To investigate the effects and mechanisms of baicalein on the proliferation and apoptosis of cisplatin-resistant gastric adenocarcinoma cell line(AGS/DDP).Methods AGS/DDP cells were treated with different concentrations of baicalein for 48 hours.The inhibition rate of cell proliferation was detected by Cell Counting Kit-8(CCK-8)assay.The apoptosis rate was determined by flow cytometry.The intracellular ATP content and mitochondrial membrane potential(MMP)were measured.The morphological changes of mitochondria in cultured gastric cancer cells were observed under electron microscopy.The mRNA expression levels of optic atrophy protein 1(OPA1)and mitochondrial metalloproteinase OMA1 in gastric can-cer cells were detected by Real-time fluorescence quantitative reverse transcription polymerase chain reaction(RT-qPCR).Results Compared with the control group,baicalein significantly inhibited the proliferation and promoted the apoptosis of AGS/DDP cells,as well as decreased the ATP level and mitochondrial membrane potential(P<0.05).Compared with AGS cells,AGS/DDP cells showed a long spindle shape and a significant trend of mitochondrial fusion,while the mRNA expression of OPA1 was significantly increased and that of OMA1 was significantly decreased(P<0.001).Conclusion Baicalein can inhibit the proliferation and promote the apoptosis of drug-resistant gastric cancer possibly cells through the mitochondrial pathway.
