仙灵骨葆胶囊组方药物致绝经后骨质疏松大鼠模型肝损伤风险的研究
Analyzing the risk of liver injury in a rat model of postmenopausal osteoporosis induced by medicinals of Xianling Gubao Capsule(仙灵骨葆胶囊)

Abstract
Objective To evaluate the differences in liver injury in postmenopausal osteoporosis(PMOP)model rats caused by various medicinals in the prescription of Xianling Gubao Capsule(仙灵骨葆胶囊,XLGBC),analyze the toxicity of main medicinals and the tox-icity-reducing effect via compatibilit,and preliminarily explore the mechanisms of toxicity.Methods A PMOP rat model was established by the method of bilateral ovary removal,and the animals were grouped according to the theory of XLGBC,and the drug was gavaged for 28 days.During the period of administration,the general condition and body mass changes of rats in each group were observed.At the end of administration,liver function-related serum biochemical indexes,organs coefficients,liver histopathology and indexes related to oxidative stress of liver tissues were examined.Results ALT was significantly elevated in the Epimedii Folium(EF)+Psoraleae Fructus(PF)group compared to the model group(P<0.01),and steatosis was the most serious in the EF+PF group.Compared with the EF+PF group,ALT was significantly lower in the EF+PF+Dipsaci Radix(DR)+Salviae Miltiorrhizae Radix Et Rhizoma(SR)group,EF+PF+Rehmannia glutinosa Libosch.(RL)+asphodeloides Bge.(AR)group,EF+PF+DR group,EF+PF+SR group,EF+PF+RL group,and EF+PF+AR group(P<0.05);and the EF+PF+SR group had the least degree of hepatic steatosis.Compared with the model group,SOD was significantly reduced in the EF+PF group(P<0.01);compared with the EF+PF group,SOD was sig-nificantly elevated in the whole formula group and the EF+PF+SR group(P<0.05);GSH was significantly elevated in the whole for-mula group,EF+PF+DR+SR group,EF+PF+RL+AR group,EF+PF+DR group,EF+PF+SR group(P<0.05);MDA was significantly reduced in the EF+PF+RL+AR group,EF+PF+DR group,EF+PF+SR group(P<0.05).Conclusion EF and PF were the main medicinals contributing to the risk of liver injury in XLGBC,and the risk of liver injury can be effectively reduced with compatibility of the formula,in which simultaneous combination of Salvia miltiorrhiza showed the strongest effect in reducing toxicity,and the mechanism of injury and attenuation may be related to oxidative stress in hepatocytes.关键词
