Exploring the mechanism of Er'zhi Pill(二至丸)in delaying aging based on gut microbiota and metabolomics

Abstract
Objective To analyze the mechanism of Er'zhi Pill(二至丸,EZP)in delaying aging by combining gut microbiota and metabolomics.Methods A natural aging rat model was established and intervened with EZP-containing feed.Enzyme-linked immu-nosorbent assay(ELISA)was used to detect changes in superoxide dismutase(SOD)and malondialdehyde(MDA).Hepatic and renal tissues were observed using hematoxylin-eosin(HE)staining.16S rDNA sequencing technology was utilized to explore the changes in gut microbiota.Ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS)was employed for metabolomics analysis.Spearman correlation analysis was conducted to investigate the correlation between metabolites and gut microbiota.Results Compared with the natural aging model group,the MDA content in the medium-dose and low-dose groups of EZP was significantly reduced(P<0.05),and no significant pathological changes were observed in the liver and kidney tissues.The 16S rDNA results showed that the relative abundance of the beneficial bacteria such as Blautia,Faecalibaculum,and Lactobacillus was increased by EZP.Identified by UPLC-Q-TOF-MS,the 13 differential metabolites related to aging were mainly enriched in 19 path-ways such as arachidonic acid(AA)metabolism and glycerophospholipid(GP)metabolism.Spearman analysis showed that some metab-olites were significantly correlated with specific gut microbiota.Conclusion EZP may delay aging by improving gut microbiota composi-tion and influencing AA and GP metabolisms.关键词
