Exploring the oxidative stress and mitochondrial protection mechanism of Shenling Baizhu Powder(参苓白术散)in alleviating ulcerative colitis based on the KEAP1/NRF2/ARE path-way

Abstract
Objective To explore the mechanism of oxidative stress and mitochondrial protection of Shenling Baizhu Powder(参苓白术散,SLBZP)in alleviating ulcerative colitis(UC)based on the KEAP1/NRF2/ARE signaling pathway.Methods Seventy-two SD rats were randomly divided into the control group(n=12)and modeling group(n=60).After successful construction of the UC modeling induced by TNBS,the rats were randomly divided into 5 groups,with 12 rats in each,and given corresponding drug treatments for 21 days.Disease activity index(DAI)scores,colonic mucosal damage index(CMDI)scores,serum inflammatory factor levels,colonic histological damage index(TDI)scores,oxidative stress indicators,pathological changes,mitochondrial ultrastructure and protein ex-pression were compared among the rats in each group.Results Compared with the control group,DAI,CMDI,TDI scores,serum IL-1β,IL-6,TNF-α,MPO levels were increased in the TNBS group(P<0.01).There was ulcer formation in the colonic mucosa and a large number of inflammatory cells infiltrated in the submucosal layer;the number of mitochondria was reduced,and the structure of the mitochondria was disrupted;the ROS levels in the colonic tissues were increased;GSH-PX and CAT levels were reduced;Nrf2,ARE and MFN2 protein expression was reduced;Keap-1 and NF-κB protein expression was elevated(P<0.01).Compared with the TNBS group,DAI,CMDI and TDI scores,serum IL-1β,IL-6,TNF-α and MPO levels were reduced in the administered groups(P<0.01);colonic mucosal ulceration was alleviated and inflammatory cell infiltration was reduced;mitochondrial number and structure were improved;ROS levels in the colonic tissues were reduced;GSH-PX,CAT levels were elevated;Nrf2,ARE,MFN2 and MFN2 protein expressions were increased;Keap-1 and NF-κB protein expressions were decreased(P<0.05).Conclusion SLBZP protects against colonic mucosal injury in UC rats by promoting Keap-1/Nrf2/ARE pathway activation and inhibiting oxidative stress.关键词
