摘要

Abstract

Objective To investigate the intervention effect and mechanism of Qingzao Jiufei Decoction(清燥救肺汤,QZJFD)on pul-monary fibrosis in mice.Methods Seventy-two SPF-grade male C57BL/6J mice were randomly divided into six groups(n=12 per group):the control group(Con),model group(M),low/medium/high-dose QZD(QZDL,QZDM,QZDH),and prednisone group(P).A pulmonary fibrosis model was established via intratracheal injection of bleomycin(5mg/kg).Serum and lung tissues were col-lected for analysis.Pathological changes were observed using hematoxylin-eosin(HE)and Masson staining.ELISA was performed to measure levels of IL-1β,TNF-α,IL-18,IL-6,α-SMA,and Col I.Biochemical assays was used to detect LDH,SOD,and CAT expressions,while DHE staining to measurereactive oxygen species(ROS).Immunohistochemistry(IHC)was used to evaluateP16,P21,and NLRP3 positive expressions,and Western blot(WB)to analyze NLRP3,Caspase-1,ASC,P16,and P21 protein expres-sions.Results Compared with the Con group,the M group exhibited diffuse inflammatory cell exudation,infiltration,and blue collagen deposition in the lung tissues.These pathological changes were alleviated to varying degrees in the QZDL,QZDM,QZDH,and P groups.The QZDM,QZDH,and P groups showed significant reductions in IL-6,IL-1β,TNF-α,IL-18,α-SMA,and Col Ⅰ lev-els(P<0.01 or P<0.05),whereas the QZDL group showed no significant decrease in IL-1β and TNF-α(P>0.05).ROS and LDH levels were significantly decreased(P<0.01 or P<0.05),while SOD and CAT levels were markedly increased(P<0.01)in the QZDL,QZDM,QZDH,and P groups.Additionally,P21,P16,and NLRP3 positive expressions were decreased,and NLRP3,ASC,P21,and P1 6 protein expressions were significantly down-regulated(P<0.01).Caspase-1 expression showed no significant differ-ence among groups(P>0.05).Conclusion QZJFD may exert anti-fibrotic effects by modulating the NLRP3/IL-1β signaling path-way,inhibiting cellular senescence,and alleviating oxidative stress.
