Abstract

Objective To analyze the action mechanism of Taohong Siwu Decoction(桃红四物汤,THSWD)in relieving lower extremity arteriosclerosis obliterans(ASO)in mice by inhibiting the TLR4/MyD88/NF-κB signaling pathway.Methods Twelve male C57BL/6 mice were selected as the blank group,and 48 8-week male ApoE mice were randomly divided into the model group,low-dose group,medium-dose group,and high-dose group,with 12 mice in each group.The blank group was fed with a normal diet,while the other groups were fed with a high-fat diet.Starting from the 9th week,the mice were gavaged;the blank group and model group received 0.2 mL of normal saline(NS)daily,while the remaining groups received0.2 mL of THSWD daily.After converting doses between humans and mice,the final dosages for various groups were approximately 0.06 g/day for the low-dose group,0.13 g/day for the medium-dose group,and 0.26 g/day for the high-dose group,with a consecutive 12 weeks of gavage.Hematoxylin-eosin(HE)staining was used to observe the morphological changes in the arterial vessels of mice;an automatic biochemical analyzer to measure the blood lipid levels in the mice;Western blot(WB)was used to detect the expressions of TLR4,MYD88,p65,p-p65,IKB,and p-pIKB in the vascular tissues.Results Compared with the blank group,mice in the model group,low-dose group,medium-dose group,and high-dose group showed significant formation of atherosclerotic plaques,while the atherosclerosis degree was significantly reduced in the low-dose,medium-dose,and high-dose groups compared to the model group.Compared with the blank group,LDL,CHO,and TG levels in the other groups were significantly elevated,while HDL level was decreased.After intervening with THSWD,the LDL,CHO,and TG levels in the low-dose,medium-dose,and high-dose groups were significantly reduced compared to the model group,while HDL levels significantly increased(P<0.05).Compared with the blank group,the expressions of TLR4,MYD88,p-IκB,and p-P65 in the other groups were significantly increased.After intervening with THSWD,the expressions of TLR4,MYD88,p-IκB,and p-P65 in the low-dose,medium-dose,and high-dose groups were significantly reduced compared to the model group(P<0.05).Conclusion THSWD can lower blood lipid levels in mice and inhibit the progression of atherosclerosis,and its mechanism of action may be related to the inhibition of the TLR4/MyD88/NF-κB signaling pathway.
