丹栀逍遥散调控Beclin-1相关自噬诱导乳腺癌细胞铁死亡的机制研究
Study on the mechanism of Danzhi Xiaoyao San(丹栀逍遥散)inducing Ferroptosis in Breast Cancer cells by regulating Beclin-1 related autophagy

Abstract
Objective To investigate the regulatory effect and mechanism of serum containing Danzhi Xiaoyao San(丹栀逍遥散DZXY)on iron death in breast cancer MDA-MB-231 cells.Methods MDA-MB-231 cells were divided into six groups:blank control(NC),blank serum(Ctrl),Danzhi Xiaoyao San(DZXY),iron death inducer(SAS),iron death inhibitor(Fer-1),and Fer-1+DZXY group.The proliferation rate and cell death were assessed using the MTT assay and Annexin V/PI double staining method,respec-tively.Mitochondrial morphology was observed by transmission electron microscopy.The levels of malondialdehyde(MDA),superoxide dismutase(SOD),ferrous ions(Fe2+),and glutathione(GSH)were measured using specific kits.The expression of glutathione perox-idase 4(GPX4),ferritin heavy chain(FTH1),transferrin receptor(TFR1),ferroportin(FPN1),ferroptosis suppressor protein 1(FSP1),Beclin-1 gene(BECN1),and microtubule-associated protein 1 light chain 3(LC3)were detected by Western blot.Results Compared to the Ctrl group,the DZXY group showed a decreased proliferation rate(P<0.01)and an increased number of dead cells(P<0.01).The levels of MDA and Fe2+increased(P<0.01,P<0.05),while SOD and GSH levels decreased(P<0.01,P<0.05).The protein expressions of TFR1,Beclin-1,and LC3-Ⅱ/Ⅰ were elevated(P<0.01,P<0.05),whereas the expressions of GPX4,FTH1,FPN1,and FSP1 were reduced(P<0.01,P<0.05).The Fer-1 group exhibited opposite trends in all indicators,while the combination of Fer-1 and DZXY could partially reverse these changes.Additionally,transmission electron microscopy re-vealed mitochondrial shrinkage and other morphological changes following drug intervention.Conclusion Serum containing Dan-Zhi-Xiao-Yao-San may promote iron death in MDA-MB-231 cells by regulating Beclin-1-mediated autophagy.关键词
