淡豆豉抑制ACE作用及其活性成分初步研究
A preliminary study on the ACE inhibitory effect and active components of Sojae Semen Pra-eparatum

Abstract
Objective To investigate the inhibitory effect of Sojae Semen Praeparatum(SSP)on angiotensin-converting enzyme(ACE)and its active components.Methods Samples were collected at different time points during SSP processing following a previously estab-lished laboratory procedure.High-performance liquid chromatography(HPLC)and ultra-performance liquid chromatography/tandem mass spectrometry(UPLC-MS/MS)were used to quantify daidzein,genistein,and γ-aminobutyric acid(GABA)in each sample.The Cushman method was modified to determine ACE inhibition,using hippuryl-histidyl-leucine(HHL)as the substrate and measur-ing absorbance with a UV spectrophotometer.The correlation between the contents of daidzein,genistein,GABA,and ACE inhibition rate was analyzed using SPSS statistical software at each time point in the process.Results The contents of daidzein and genistein peaked at(0.951±0.296)mg/g and(0.920±0.170)mg/g,respectively,after 12 days of additional fermentation,while GABA content reached itshighestlevelof(7.03±1.29)mg/gafter15 days.The ethanol extracts from different time points exhibited low and relatively stable ACE inhibition(r<0,P>0.05),whereas the water extracts showed significant ACE inhibitory activity,with a maximum inhibi-tion rate of 90.62%.A strong positive correlation was observed between GABA content and ACE inhibition(r>0.99,P<0.05).Con-clusion SSP exhibits potent ACE inhibitory activity in vitro,and GABA is likely the key active component responsible for this effect.关键词
