桃红四物汤改善大鼠腹部皮瓣术后缺血再灌注损伤JNK信号通路的机制研究
Mechanism study of Taohong Siwu Decoction(桃红四物汤)in ameliorating ischemia-reper-fusion injury in rats of abdominal flaps post-surgery based on the JNK signaling pathway

Abstract
Objective To investigate the effect and mechanism of Taohong Siwu Decoction(桃红四物汤,THSWD)on ischemia-reperfusion injury(IRI)in rats after abdominal flaps surgery.Methods Thirty-two rats were randomly divided into the blank group,model group,JNK inhibitor group(SP600125),and THSWD group.The blank group underwent a sham surgery,while the other groups established an abdominal flap IRI model.Interventions were administered for 7 days postoperatively.Flap morphology and pathological changes were observed,while the levels of p-ASK1,p-JNK,Bax proteins,and serum inflammatory factors were measured.Results In the THSWD group,flap edema and inflammatory exudation were significantly reduced in flaps post-surgery.HE staining showed mil-der tissue edema and fewer inflammatory cells.Western blot(WB)analysis revealed significant differences in the expressions of Bax,p-ASK1,and p-JNK proteins in the JNK inhibitor group compared to the model group(P<0.01,P<0.01,P<0.05).The THSWD group demonstrated significantly lower expressions of these proteins compared to the JNK pathway inhibitor group(P<0.001).ELISA results indicated that both the JNK inhibitor and THSWD groups had significantly lower expressions of serum inflammatory factors TNF-α,IL-1β,and IL-6 than the model group(P<0.0001).No statistical differences were found in inflammatory factor expressions be-tween the JNK inhibitor and THSWD groups(P>0.05).Conclusion THSWD alleviates flap IRI by regulating the JNK signaling path-way,inhibiting apoptosis,and reducing postoperative inflammatory response.关键词
