基于TXNIP/NLRP3信号通路探讨化浊解毒方对5/6肾切除大鼠肾脏纤维化的影响
Exploring the effect of Huazhuo Jiedu Formula(化浊解毒方)on renal fibrosis in 5/6 nephrec-tomy rats based on TXNIP/NLRP3 signaling pathway

Abstract
Objective To investigate the mechanism by which Huazhuo Jiedu Formula(化浊解毒方,HZJDF)ameliorates renal fibrosis in5/6 nephrectomized rats through the TXNIP/NLRP3 pathway.Methods Sixty-seven SD rats were divided into the sham group(n=11)and modeling group(n=56).The latter underwent5/6 nephrectomy and were further divided into the model group,low/medium/high-dose HZJDF groups(1.175 g·kg-1,2.35 g·kg-1,4.7 g·kg-1),and Losartan Potassium group(0.0045 g·kg-1)for 8-week intervention.Serum levels of IL-1β and IL-18 were measured.Renal histopathology was assessed by HE and Masson staining.Western blot(WB)was used to detect protein expression of TGF-β1,PDGF-B,CTGF,α-SMA,Collagen-I,TXNIP,NLRP3,and Caspase-1.Immunohistochemistry(IHC)was used to analyze the positive expression of TXNIP,NLRP3,and Caspase-1.Results Compared with the sham group,the model group showed significantly elevated IL-1β,IL-18,fibrosis-and pyroptosis-related pro-teins(P<0.01),along with severe renal injury.The medium/high-dose HZJDF and losartan groups showed reduced inflammatory fac-tors and fibrosis markers(P<0.05 or P<0.01),suppressed TXNIP,NLRP3,Caspase-1 expression(P<0.05 or P<0.01),and alleviated pathological damage,while the low-dose HZJDF group showed improvement in only partial indicators.Conclusion HZJDF may reduce pyroptosis,mitigate inflammation and fibrosis,and protect renal function by inhibiting the TXNIP/NLRP3 pathway.关键词
