连朴饮对糖尿病胃轻瘫大鼠Cajal间质细胞及巨噬细胞极化的影响
Effect of Lianpo Decoction(连朴饮)on ICC and macrophage polarization in diabetic gastrop-aresis rats

Abstract
Objective To observe the effect of Lianpo Decoction(连朴饮,LPD)on interstitial cells of Cajal(ICC)and macrophage po-larization in rats with diabetic gastroparesis(DGP).Methods The low/medium/high-dose LPD groups received 4.9,9.8,and 19.6 g·kg-1·d-1via gavage,respectively,while the domperidone group received 3.15 mg·kg-1·d-1 by gavage.All treatments lasted 8 weeks.After 8 weeks,general conditions were observed,and gastric emptying rate and small intestinal propulsion rate were measured.ICC quantity and morphology were examined under electron microscopy.CD86 and CD206 protein expressions in gastric antrum tissues were detected via immunohistochemistry(IHC),and serum superoxide dismutase(SOD)was measured using the hydroxylamine meth-od.Serum malondialdehyde(MDA)was assessed via the thiobarbituric acid(TBA)method,while serum interleukin-1β(IL-1β)and interleukin-10(IL-10)contents were quantified via ELISA.Protein expressions of c-Kit and heme oxygenase-1(HO-1)in gastric antrum tissues were analyzed via Western blot(WB),and mRNA expressions of c-Kit and HO-1 were detected via RT-qPCR.Results Compared with the blank group,the model group showed significantly reduced ICC quantity in the gastric antrum,accom-panied by mitochondrial autophagy,cytoplasmic vacuolation,Golgi apparatus fragmentation,and cristae dissolution.Gastric emptying and small intestinal propulsion rates were significantly reduced(P<0.05).IL-1β,MDA levels,and CD86 protein expression levels were significantly increased(P<0.05),while IL-10,SOD levels,CD206 protein level,and c-Kit and HO-1 protein/mRNA levels were significantly decreased(P<0.05).Compared with the model group,the low/medium/high-dose LPD groups and the domperi-done group exhibited increased ICC count,ameliorated morphology,elevated gastric emptying and small intestinal propulsion rates(P<0.05).The contents of IL-1β and MDA and CD86 protein expression level were significantly decreased(P<0.05),while the contents of IL-10 and SOD,CD206 and protein expression levels were significantly increased(P<0.05).The medium/high-dose LPD groups and domperidone group showed significantly increased c-Kit,HO-1 protein and mRNA levels(P<0.05).Conclusion LPD allevi-ates DGP by regulating oxidative stress,mediating macrophage polarization,and restoring ICC quantity and function.关键词
